The aim of the study was to investigate the morphological and metabolic features of primary hippocampal cultures formation on hydrogel films and scaffolds based on hyaluronic acid glycidyl methacrylate. Materials and Methods. Hydrogel films and scaffolds with certain architectonics were developed by micromolding technique on the basis of hyaluronic acid glycidyl methacrylate. Primary hippocampal cells obtained from C57BL/6 mouse embryos (E18) were cultured on the created constructs more than 14 days. Testing cell viability, morphometric assessment, and analysis of spontaneous calcium activity of primary hippocampal cultures were performed on day 14 of cultures development in vitro. Results. This study revealed that the material for the development of scaffolds with given architectonics is non-toxic for the nervous system cells. Dissociated hippocampal cells were actively attached to the scaffold surface and were assembled into cell conglomerates, which exhibited spontaneous calcium activity. Conclusion. Scaffolds designed on the basis of hyaluronic acid glycidyl methacrylate have a high biocompatibility with the nervous system cells. Architectonics and adhesive properties of scaffold contribute to the formation of functionally active cell conglomerates.

To evaluate the efficiency and safety of the contrast agent based on HSA-coated MNP on the model of the C6 glioma. Materials and methods. Extensive preclinical research of specific activity has been performed. Studies were conducted on the model of the orthotopic C6 glioma. Results. In the preclinical studies, it was demonstrated that HSA-coated magnetic iron oxide nanoparticles (MNP-HSA) accumulate in the tumor and its large vessels. Conclusion. Magnetic resonance imaging with contrast agent allows visualization of the tumor tissue and its vascularization.

We have developed a method of the terahertz (THz) solid immersion microscopy for the reflection-mode imaging of soft biological tissues. It relies on the use of the solid immersion lens (SIL), which employs the electromagnetic wave focusing into the evanescent-field volume (i.e. at a small distance behind the medium possessing high refractive index) and yields reduction in the dimensions of the THz beam caustic. We have assembled an experimental setup using a backward-wave oscillator, as a source of the continuous-wave THz radiation featuring λ= 500 μm, a Golay cell, as a detector of the THz wave intensity, and a THz SIL comprised of a wide-aperture aspherical singlet, a truncated sphere and a thin scanning windows. The truncated sphere and the scanning window are made of high-resistivity float-zone silicon and form a unitary optical element mounted in front of the object plane for the resolution enhancement. The truncated sphere is rigidly fixed, while the scanning window moves in lateral directions, allowing for handling and visualizing the soft tissues. We have applied the experimental setup for imaging of a razor blade to demonstrate the advanced 0:2λ resolution of the proposed imaging arrangement. Finally, we have performed imaging of sub-wavelength-scale tissue spheroids to highlight potential of the THz solid immersion microscopy in biology and medicine.

© 2018 Bentham Science Publishers. A number of biological and clinical characteristics typical of late life depression (LLD) have been suggested by recent research findings. The close association of LLD with cognitive impairment is now well documented and evidenced. However, it is still not clear whether it is depression that leads to cognitive decline, and in more severe cases, to dementia. The work presented in this review article suggests that depression and dementia frequently and strongly copresent, even if the causality remains largely opaque.

В области изучения нейробиологии униполярных депрессивных расстройств (УДР) перспективными считаются нейровизуализационные методы, особенно позитронно-эмиссионная томография (ПЭТ) и функциональная магнитно-резонансная томография (фМРТ). В статье приводится обзор современных нейровизуализационных данных, касающихся структурно-функциональных особенностей головного мозга у лиц, страдающих УДР. Результаты отдельных исследований представлены в зависимости от особенностей методов их проведения (состояние покоя, выполнение когнитивных и эмоциональных тестов) и соотнесены с основными нейробиологическими концепциями развития депрессивных расстройств. Отдельно рассмотрены возможности нейровизуализационных исследований для оценки и прогнозирования результатов антидепрессивной терапии.

Objectives: Central (hypogonadotropic) hypogonadism in women could be a cause of persistent amenorrhea and hypoestrogenemia as observed in postmenopause. This study aimed to compare the clinical, hormonal and biochemical features in women with non-physiological (central hypogonadism) and physiological (postmenopause) hypoestrogenemia.
Methods: A total of 161 young women, median age 24.9 years (interquartile range (IQR) 21.2; 30.5) with central hypogonadism (with isolated hypogonadotropic hypogonadism, n = 76, and with hypopituitarism, n = 85), 53 healthy young women, median age 23.9 years (IQR 23.1; 28.0) and 50 healthy postmenopausal women, median age 56.0 years (IQR 53.1; 58.5), were examined. Psychoemotional, neurovegetative and urogenital symptoms, sex steroid levels, parameters of lipid and mineral metabolism were evaluated.
Results: In young women with central hypogonadism, the frequencies of psychoemotional, neurovegetative and urogenital complaints differed significantly from those in healthy young women and were similar to those in postmenopausal women. Concentrations of estradiol, testosterone, dehydroepiandrosterone sulfate, parameters of lipid and mineral metabolism as well as quality of life in women with central hypogonadism were not typical of healthy young women but were similar to those of postmenopausal women of middle/old age.
Conclusions: Despite the young age of women with central hypogonadism, clinical, hormonal and biochemical abnormalities were similar in many aspects to those in postmenopausal women at middle/old age. These revealed features could be considered as signs of premature aging in young amenorrheic women with low gonadotropin levels.

Studies on the methods of nanoparticle (NP) synthesis, analysis of their characteristics, and exploration of new fields of their applications are at the forefront of modern nanotechnology. The possibility of engineering water-soluble NPs has paved the way to their use in various basic and applied biomedical researches. At present, NPs are used in diagnosis for imaging of numerous molecular markers of genetic and autoimmune diseases, malignant tumors, and many other disorders. NPs are also used for targeted delivery of drugs to tissues and organs, with controllable parameters of drug release and accumulation. In addition, there are examples of the use of NPs as active components, e.g., photosensitizers in photodynamic therapy and in hyperthermic tumor destruction through NP incorporation and heating. However, a high toxicity of NPs for living organisms is a strong limiting factor that hinders their use in vivo. Current studies on toxic effects of NPs aimed at identifying the targets and mechanisms of their harmful effects are carried out in cell culture models; studies on the patterns of NP transport, accumulation, degradation, and elimination, in animal models. This review systematizes and summarizes available data on how the mechanisms of NP toxicity for living systems are related to their physical and chemical properties.
