© 2020 Elsevier B.V. Brain metastases manifest the advanced stage of breast cancer disease with poor prognosis for patient survival. Recent reports demonstrate that some therapeutic agents can activate the expression of several breast cancer-associated genes, whose products are involved in the onset and development of brain metastases. In this study, we discovered a functional link between KISS1 and E-cadherin that could be observed in both primary brain metastatic lesions and paired cell lines, such as parental CN34TGL and MDA-MB-231 and their respective brain metastatic subclones CN34Brm2Ctgl and MDA-MB-231Br. Remarkably, expression of KISS1 and E-cadherin genes consistently showed an inverse correlation in all of the above cell/tissue types. While E-cadherin expression was strongly upregulated in metastatic clones isolated from blood and brain, the levels of this protein in parental MDA-MB-231 cell line was low. Furthermore, E-cadherin upregulation can be artificially induced in MDA-MB-231Br and CN34Brm2Ctgl cell populations by knocking down KISS1 expression directly or through overexpressing the miR345 mimic. In the aggregate, our data suggest that the tumor microenvironment, which controls breast cancer spreading via miR345-regulated KISS1 expression, might modulate metastatic spreading by a mechanism(s) involving upregulation of E-cadherin production.

© 2020 This work is dedicated to the influence of thermal treatment procedures in vacuum and air atmosphere on the structural features of multiwalled TiO2 anodic nanotubes (NTs) formed in fluorine-containing ethylene glycol (EG) based electrolyte investigated by high-resolution transmission electron microscopy (HRTEM) in conjunction with EDX and electronic nanodiffraction (STEM nanodiffraction), IR and Raman spectroscopy, XPS and ToF SIMS. Using electron nanodiffraction technique, there were estimated TiO2 nanocrystallite sizes for the inner layer of the NTs after annealing in air and vacuum for the first time. The differences in carbon distribution profiles in TiO2 nanotube crosscut after thermal treatments in air and vacuum were discussed. It was found that thermal treatment in vacuum leads to different phase composition of TiO2 NTs IL comparing to annealing in air. A sequential thermal treatment (firstly in vacuum and then in air) enables to remove carbon from TiO2 NTs surface and partly from TiO2 NTs IL and prevent detachment of IL from OL at the same time. The obtained data will be useful for understanding the mechanisms of structural modifications during thermal treatments as well as for functional properties optimization of such nanostructures.

In Video 1 we illustrate our technique for the anterior ITF approach for the surgical treatment of a middle cranial fossa meningioma invading the ITF. This surgical video describes the anterior ITF approach in 2 steps. First, a standard extradural middle fossa approach subtemporal approach is performed on a cadaveric specimen, illustrating the anterior extension to the cavernous sinus. Second, the anterior ITF approach is performed for the surgical treatment of a temporal lobe meningioma with extension to the anterior ITF. This technique provides a minimally invasive approach for treating middle fossa lesions with anterior ITF extension.

CDH13 is involved in neurite outgrowth and axon guidance during early brain development and we previously provided evidence that constitutive CDH13 deficiency influences the formation of the raphe serotonin (5-HT) system by modifying neuron-radial glia interaction. Here, we dissect the specific impact of CDH13 on 5-HT system development and function using a 5-HT neuron-specific Cdh13 knockout mouse model (conditional Cdh13 knockout, Cdh13 cKO). Our results show that exclusive inactivation of CDH13 in 5-HT neurons selectively increases 5-HT neuron density in the embryonic dorsal raphe, with persistence into adulthood, and serotonergic innervation of the developing prefrontal cortex. At the behavioral level, adult Cdh13 cKO mice display delayed acquisition of several learning tasks and a subtle impulsive-like phenotype, with decreased latency in a sociability paradigm alongside with deficits in visuospatial memory. Anxiety-related traits were not observed in Cdh13 cKO mice. Our findings further support the critical role of CDH13 in the development of dorsal raphe 5-HT circuitries, a mechanism that may underlie specific clinical features observed in neurodevelopmental disorders.

© 2020 Elsevier Ltd The 1986 accident at the Chernobyl Nuclear Power Plant led to large-scale changes in the environmental situation. The purpose of our study was to conduct a comparative analysis of the morphological states of the dentition of individuals living in regions exposed to radiation to determine the groups at risk for the main classes of dental anomalies. We believe our results will support the development of a differentiated system for dental rehabilitation and follow-up of individuals exposed to radiation. The prevalence rate of dental anomalies was studied in 1,889 patients of both sexes divided by age in accordance with dentition formation stages and by regions of residence in accordance with the 137Cs soil-contamination level. A statistically significant decrease was observed in the number of patients with normal dentition for their age among those who had been exposed to prenatal radiation. A sharp increase in combined dental anomalies was revealed in patients who lived in regions with a137Cs soil-contamination level ranging from 555 to 1665 GBq/km2; concomitantly, multidirectional fluctuations were observed in the numbers of tooth and occlusion anomalies. Among the examined population, the most severe pathology of the oral organs was found in prenatally irradiated patients (born between April 26, 1986, and April 30, 1987). The prevalence of dental anomalies is interrelated not only with the level of radioactive contamination in the soil of the dwelling area, but also with the age of the surveyed individuals at the moment of the accident.

© 2020 Elsevier B.V. The development of a highly efficient and rapid method for the accurate preparation of photocatalysts with novel morphologies is a hot research topic. The different morphologies of BiVO4 was prepared using surfactants-assisted microwave method, and demonstrated irregular (no surfactant), octahedral (sodium dodecyl benzene sulfonate), olive-like (polyvinylpyrrolidone) and hollow structures (ethylenediaminetetraacetic acid), respectively. The BiVO4-CdS were synthesized using the chemical-bath-deposition method with different morphologies of BiVO4 as the substrates. The hollow structure of BiVO4 displayed the highest photocatalytic performance. Moreover, the photodegradation rates of the hollow structure BiVO4-CdS on tetracycline hydrochloride and ciprofloxacin were about 1.8 and 1.5 times higher than the corresponding BiVO4, indicating that the Z-scheme heterojunction can improve the photogenerated electron pairs separation efficiency. Furthermore, the regulation mechanism of morphology and energy-band position, as produced using the surfactants, has also been thoroughly investigated in this work, which provides a novel insight into the efficient and rapid preparation of photocatalysts with special morphology and high performance.

The present study presents an attempt to modify the surface properties of macroporous resins (MRs) in order to improve anthocyanin adsorption and desorption from Pyrus communis var Starkrimson fruit peel extract. A number of MRs were tested to optimise the ultrasonic-assisted adsorption (UAA) conditions; including ultrasonic power (100–400 W), resin-to-extract ratio (1–3 g/50 mL) and temperature (20–40 °C). Similarly, varying ultrasonic-assisted desorption (UAD) conditions were optimised; including ultrasonic power (200–600 W), resin-to-solvent ratio (1–4 g/50 mL), ethanol concentration (60–90% v/v) and temperature (20–40 °C). The Amberlyst 15 (H) cationic resin was found to be superior to the other tested resins. The maximum adsorption capacity (659 µg/g) of cyanidin 3-galactoside (Cy 3-gal) was achieved under the optimised UAA conditions (400 W, 20 °C and 1 g/50 mL), while 616 µg/g of Cy 3-gal was recovered under the optimised UAD conditions (582 W, 1 g/50 mL, 60% and 20 °C). Moreover, titratable-acid and total-sugar contents were found to be significantly lower under UAA than under conventional-assisted adsorption (CAA). ANOVA revealed that process factors had significant effects on the Cy 3-gal purification, as depicted by their linear, quadratic and interactive effects. While anthocyanin adsorption was found to be significantly improved at lower ultrasonic power, higher power promoted the desorption process. Adsorption under optimized UAA conditions followed pseudo second-order kinetics and multilayer adsorption (Freundlich isotherm) onto the Amberlyst 15 (H) resin surface was observed. The particle-size distribution curve and scanning electron microscopic images also revealed higher resin-surface roughness, peeling and the appearance of pores on the surface under ultrasonication. This is the first study to use ultrasonication to modify a cationic exchange resin for the improvement of Cy 3-gal purification from a fruit extract. This study can recommend the use of ultrasonication as a low-cost green technique that can improve macroporous resin characteristics for better purification of compounds from an extract.

© 2020 Elsevier B.V. Herein four modified polymers were prepared from readily available addition poly(5-vinyl-2-norbornene) (PVNB) and their gas-transport properties were studied in detail. Hydrogenation, epoxidation, cyclopropanation and thiol-en reactions were chosen for the modifications of PVNB. Hydrogenation of PVNB was performed using p-toluenesulfonyl hydrazide. Epoxidation of PVNB was realized employing m-chloroperoxybenzoic acid. Cyclopropanation of PVNB was carried out using diazomethane in the presence of a Pd-catalyst. For thiol-en reaction, thioacetic acid was applied as the source of a thiol and AIBN as an initiator. All listed modifications were performed in high yields (≥80%) without the destruction of polymer main chains. The degree of functionalizations was up to 99%. The influence of these modifications on the properties of the resulting polymers was evaluated. Cyclopropanation and hydrogenation of PVNB led to an enhancement of gas permeability with minimal decrease in selectivity, while epoxidation or thioacetylation gave a substantial increase in CO2/N2 selectivity with decrease in permeability. The modified polymers with polar side-groups exhibited attractive selectivities for CO2/N2, CO2/CH4 and H2/N2 gas separations.

© 2019 Elsevier B.V. Background: Skeletal muscle cells continuously generate reactive oxygen species (ROS). Excessive ROS can affect lipids resulting in lipid peroxidation (LPO). Here we investigated the effects of myotube intracellular calcium-induced signaling eliciting contractions on the LPO induction and the impact of LPO-product 4-hydroxynonenal (4-HNE) on physiology/pathology of myotubes using C2C12 myoblasts. Methods: C2C12 myoblasts were differentiated into myotubes, stimulated with caffeine and analyzed for the induction of LPO and formation of 4-HNE protein adducts. Further effects of 4-HNE on mitochondrial bioenergetics, NADH level, mitochondrial density and expression of mitochondrial metabolism genes were determined. Results: Short and long-term caffeine stimulation of myotubes promoted superoxide production, LPO and formation of 4-HNE protein adducts. Furthermore, low 4-HNE concentrations had no effect on myotube viability and cellular redox homeostasis, while concentrations from 10 μM and above reduced myotube viability and significantly disrupted homeostasis. A time and dose-dependent 4-HNE effect on superoxide production and mitochondrial NADH-autofluorescence was observed. Finally, 4-HNE had strong impact on maximal respiration, spare respiratory capacity, ATP production, coupling efficiency of mitochondria and mitochondrial density. Conclusion: Data presented in this work make evident for the first time that pathological 4-HNE levels elicit damaging effects on skeletal muscle cells while acute exposure to physiological 4-HNE induces transient adaptation. General significance: This work suggests an important role of 4-HNE on the regulation of myotube's mitochondrial metabolism and cellular energy production. It further signifies the importance of skeletal muscle cells hormesis in response to acute stress in order to maintain essential biological functions.

© 2019 Background: Relatively few studies investigated the importance of frailty in radical cystectomy (RC) patients. We tested the ability of frailty, using the Johns Hopkins Adjusted Clinical Groups indicator, to predict early perioperative outcomes after RC. Methods: RC patients were identified within the National Inpatient Sample database (2000–2015). The effect of frailty, age and Charlson Comorbidity Index were tested in five separate multivariable models predicting: (1) complications, (2) failure to rescue (FTR), (3) in-hospital mortality, (4) length of stay (LOS) and (5) total hospital charges (THCs). All models were weighted and adjusted for clustering, as well as all available patient and hospital characteristics. Results: Of 23,967 RC patients, 5833 (24.3%) were frail, 7721 (32.2%) were aged ≥75 years and 2832 (11.8%) had CCI ≥2. Frailty, age ≥75 years and CCI ≥2 were non-overlapping in 86.3% of the cohort. Any two or three of these features were recorded in 12.4 and 1.3%, respectively. Frailty was an independent predictor of all five examined endpoints and the magnitude of its association was stronger or at least equal than that of age ≥75 years and CCI ≥2. Conclusion: Frailty, advanced age and comorbidities represent non-overlapping patients’ characteristics. Of those, frailty represents the most consistent and strongest predictor of early adverse outcomes after RC. Ideally, all three indicators should be considered in retrospective, as well as prospective analyses. Pre-surgical recognition of frail patients should be ideally incorporate in clinical practice in order to address these patients to multimodal pre-habilitation programs that may potentially improve the perioperative prognosis.

According to different authors, the incidence of this condition varies in severity and being recorded in 10–70% of cases during chronic intermittent hemodialysis. The development of severe chronic intradialytic hypotension in most cases is considered as a relative contraindication to kidney transplantation due to the risk of the graft dysfunction and loss in the early postoperative period. Meanwhile, there is no consensus on the lower limit of blood pressure, which would be an absolute contraindication for kidney transplantation. In addition, patients with intradialytic hypotension have the dialysis session reduced which leads to inadequate dialysis; also, they often have such complications as thrombosis of an arteriovenous fistula, as a result of which further dialysis sessions become impossible. In such cases, renal transplantation is a risky, but lifesaving operation. We present a clinical case report of kidney allotransplantation in a female patient with a history of bilateral nephrectomy, who had been suffering from severe chronic intradialytic hypotension for three years before the operation. After allograft transplantation, her blood pressure was normalized to reference values, and the graft function fully recovered.

© 2019 Stem cells and the niche in which they reside feature a complex microenvironment with tightly regulated homeostasis, cell-cell interactions and dynamic regulation of metabolism. A significant number of organoid models has been described over the last decade, yet few methodologies can enable single cell level resolution analysis of the stem cell niche metabolic demands, in real-time and without perturbing integrity. Here, we studied the redox metabolism of Lgr5-GFP intestinal organoids by two emerging microscopy approaches based on luminescence lifetime measurement – fluorescence-based FLIM for NAD(P)H, and phosphorescence-based PLIM for real-time oxygenation. We found that exposure of stem (Lgr5-GFP) and differentiated (no GFP) cells to high and low glucose concentrations resulted in measurable shifts in oxygenation and redox status. NAD(P)H-FLIM and O2-PLIM both indicated that at high ‘basal’ glucose conditions, Lgr5-GFP cells had lower activity of oxidative phosphorylation when compared with cells lacking Lgr5. However, when exposed to low (0.5 mM) glucose, stem cells utilized oxidative metabolism more dynamically than non-stem cells. The high heterogeneity of complex 3D architecture and energy production pathways of Lgr5-GFP organoids were also confirmed by the extracellular flux (XF) analysis. Our data reveals that combined analysis of NAD(P)H-FLIM and organoid oxygenation by PLIM represents promising approach for studying stem cell niche metabolism in a live readout.

© 2019 Aims: The high sugar and lipid content of the Western diet (WD) is associated with metabolic dysfunction, non-alcoholic steatohepatitis, and it is an established risk factor for neuropsychiatric disorders. Our previous studies reported negative effects of the WD on rodent emotionality, impulsivity, and sociability in adulthood. Here, we investigated the effect of the WD on motor coordination, novelty recognition, and affective behavior in mice as well as molecular and cellular endpoints in brain and peripheral tissues. Main methods: Female C57BL/6 J mice were fed the WD for three weeks and were investigated for glucose tolerance, insulin resistance, liver steatosis, and changes in motor coordination, object recognition, and despair behavior in the swim test. Lipids and liver injury markers, including aspartate-transaminase, alanine-transaminase and urea were measured in blood. Serotonin transporter (SERT) expression, the density of Iba1-positive cells and concentration of malondialdehyde were measured in brain. Key findings: WD-fed mice exhibited impaired glucose tolerance and insulin resistance, a loss of motor coordination, deficits in novel object exploration and recognition, increased helplessness, dyslipidemia, as well as signs of a non-alcoholic steatohepatitis (NASH)-like syndrome: liver steatosis and increased liver injury markers. Importantly, these changes were accompanied by decreased SERT expression, elevated numbers of microglia cells and malondialdehyde levels in, and restricted to, the prefrontal cortex. Significance: The WD induces a spectrum of behaviors that are more reminiscent of ADHD and ASD than previously recognized and suggests that, in addition to the impairment of impulsivity and sociability, the consumption of a WD might be expected to exacerbate motor dysfunction that is also known to be associated with adult ADHD and ASD.
